뽕잎과 오디 혼합분말을 첨가한 이너 뷰티 푸드(묵) 제조 및 품질평가

요약

목적

본 연구에서는 뽕잎과 오디에 포함되어 있는 생리활성 물질들 간의 시너지 효과를 이용하여 이너 뷰티 푸드(묵)를 개발하고자 뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가한 묵을 제조하여 품질특성을 평가하였다.

방법

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가한 묵의 색도, 기계적 물성, 일반성분, 무기질 함량, 생리물질 활성을 측정하였다.

결과

명도(lightness)를 나타내는 L*값은 뽕잎과 오디분말이 1:1 비율로 2% 첨가된 ML1MF1 (32.54)의 색도가 가장 낮게 나타났다. 적색도(redness)를 나타내는 a*값은 ML0MF2 (3.94)가 가장 컸으며 ML2MF0 (-5.38)가 가장 낮은 값을 나타내었다. 황색도(yellowness)를 나타내는 b*값은 ML2MF0 (8.10)가 가장 높았고 ML0MF2 (-5.25)가 가장 낮았다. 묵의 기계적 물성을 측정한 결과, 경도는 ML0MF2 (126.07)가 가장 크고 ML2MF0 (87.27)가 가장 낮았다. 검성은 ML0MF2 (60.58)가 가장 크고 ML1.5MF0.5 (47.17)가 가장 낮았다. 씹힘성은 ML0MF2 (55.11)가 가장 크고 ML1.5MF0.5 (40.82)가 가장 낮았다. 묵의 일반성분을 분석한 결과, 조지방은 ML0.5MF1.5 (1.51%)가 가장 많았으며 ML1MF1 (1.12%)이 3번째, ML2MF0 (1.07%)가 가장 적었다. 조단백질은 ML0MF2 (4.62%)가 가장 많았으며 MLMF1 (4.44%)이 2번째, ML0.5MF1.5 (2.52%)가 가장 적게 나타났다. 묵의 무기질 함량을 분석한 결과, 칼슘, 칼륨, 마그네슘 함량은 ML0.5MF1.5이 가장 많았으며 ML1MF1이 2번째 ML0MF2가 가장 적게 나타났다. 묵의 생리활성 물질을 분석 결과, GABA, 1-deoxynojirimycin (DNJ) 함량은 ML2MF0 (3.49 mg/100 g, 5.28 mg/100 g)이 가장 많았으며 ML1MF1 (2.84 mg/100 g, 3.75 mg/100 g)이 3번째, ML0MF2 (1.34 mg/100 g, 10.3 mg/100 g)가 가장 적었다. 안토시아닌과 ycanidin-3-glucoside (C3G) 함량은 ML0MF2 (75.41 mg/100 g, 95.48 mg/100 g)가 가장 많았고 ML1MF1 (19.88 mg/100 g, 21.08 mg/100 g)이 3번째, ML2MF0 (1.97 mg/100 g, 0.1 mg/100 g)가 가장 적었다.

결론

이상의 결과 뽕잎과 오디를 1:1로 혼합하여 2% 첨가한 ML1MF1이 조단백질 2번째, 조지방, 칼슘, 칼륨, 마그네슘 모두 3번째로 많이 함유되어 있었으며, 생리활성 물질인 GABA, 1-DNJ, 안토시아닌과 C3G도 3번째로 함유되어 있었다. 그러므로 뽕잎과 오디분말을 1:1로 혼합하여 첨가하였을 때 뽕잎과 오디에 함유되어 있는 생리활성물질의 시너지 효과가 가장 바람직할 것으로 판단되며 이는 향후 각종 건강 기능성 식품이나 이너 뷰티푸드로 활용되는데 있어 긍정적일 것으로 사료된다.

Abstract

Purpose

The purpose of this study was to develop an inner beauty food (Mook) using the synergistic effect between physiologically active substances contained in mulberry leaf and fruits. 2% (w/w) of each, the mulberry leaves and fruit powders were used.

Methods

The color, mechanical properties, textural characteristics, mineral content, and the physiological activity of the Mook were measured.

Results

The lightness (L*) was lowest in ML1MF1 group compared with the control group. The redness (a*) was highest in ML0MF2 and the yellowness (b*) was highest in ML2MF0 group, both compare with the control group. The textural characteristics of the Mook indicated that hardness, gumminess, and chewiness were highest in ML0MF2. The analysis of general composition of Mook showed that crude lipid was the most abundant in ML0.5MF1.5, third the most abundant in ML1MF1, the least abundant in ML2MF0. The crude protein was the most abundant in ML0MF2, second the most abundant in ML1MF1, and the least abundant in ML1.5MF0.5. Calcium, potassium, and magnesium concentrations were the highest in ML0.5MF1.5, the second highest in ML1MF1, and ML0MF2 had the lowest concentrations. As a result of analyzing the physiologically active substance in Mook, GABA, 1-deoxynojirimycin (DNJ) content was the highest in ML2MF0, MLMF1 was third and ML0MF2 had the least. The content of anthocyanin and cyanidin-3-glucoside (C3G) was the highest in ML0MF2, third highest in ML1MF1, and the least in ML2MF0.

Conclusion

ML1MF1 had the second most abundant concentrations of crude protein, calcium, potassium and magnesium and had the third most abundant concentrations of crude fat and the physiologically active substances GABA, 1-DNJ, anthocyanin, and C3G. Therefore, the synergistic effect of the physiologically active substance contained in the mulberry leaf and fruit was found to be optimal when the mulberry leaf and fruit were mixed in equal parts. This is expected to be favorable for use as various health functional foods and inner beauty foods in the future.

目的

利用桑叶和桑果实中所含的生理活性物质之间的协同作用,开发一种内在美容食品(Mook)。利用含有 2%(w/w)的桑叶和果粉混合粉末的绿豆淀粉制备Mook并评价质量。

方法

测量Mook的颜色,机械性能,一般成分,矿物质含量和生理活性。

结果

结论

ML1MF1含有第二丰富的粗蛋白,钙,钾和镁,并且具有第三丰富的粗脂肪,生理活性物质GABA,1-DNJ,花青素和C3G。因此,当桑叶和果实以相等的份数混合时,发现桑叶和果实中含有的生理活性物质的协同效应是最佳的。预计这将有利于将来用作各种保健功能食品和内在美容食品。

Introduction

이너 뷰티 푸드(inner beauty food)는 식습관과 생활습관 등을 통해 피부를 관리하는 '먹는 화장품'으로 피부 속 건강을 좋게하여 피부미용의 효과를 배가시켜 주는 제품으로 다양한 제형 및 효과를 가지며 피부에 좋은 보조식품으로 정의하고 있다(Han & Lim, 2017). 즉, 이너 뷰티 푸드는 꾸준한 섭취를 통해 건강과 피부를 가꾸어 주는 효과를 가진 식품이며 아름다움과 직결된다는 인식으로 더욱 주목 받고 있다(Kim, 2005). 특히 최근에는 내적 건강을 통한 외면의 아름다움에 대한 관심이 고조되면서 이너 뷰티 푸드에 관심이 증대하고 있으며 이에 대한 연구도 다방면에서 진행되고 있다(Lee et al., 2018; Ju et al., 2017). 이러한 이너 뷰티 푸드에 관련해 한방에서는 뽕나무를 부종이나 당뇨 치료를 위해 잎, 줄기, 뿌리껍질 및 열매 등을 다양하게 이용해 왔다(Son et al., 2014). 뽕잎에는 rutin, γ-aminobutyric acid (GABA), chlorogenic acid, β-sitosterol, β-sitosterolglycoside, campesterol 및 DNJ 등 다양한 생리활성물질이 함유되어 있다(Choi et al., 2015; Park et al., 2013). 뽕잎의 우수한 기능성을 나타내는 대표적인 지표물질로 flavonoid 계의 rutin은 모세혈관 강화작용이 있으며, 혈압 저하 물질로 알려져 있는 GABA는 녹차에 비해 약 10배 정도 높게 함유되어 있고, DNJ은 장내 탄수화물을 분해하는 효소인 α-glucosidase의 활성을 억제하여 식후 급격히 상승하는 혈당을 조절하는 것으로 알려져 있다(Park et al., 2012). 또한 뽕잎에는 β-sitosterol, campesterol, β-sitosterol glycoside, β-ecdysone 및 inosterol 등 식물성 스테롤이 다량으로 존재하여 혈중 콜레스테롤과 중성지질의 저하작용, HDL-cholesterol의 증가 및 항산화작용 등의 기능을 나타내는 것으로 알려지고 있다(Choung & Lim, 2012).
오디는 뽕나무과에 속하는 낙엽 교목인 뽕나무(Morus alba L.)의 열매로, 함유되어 있는 anthocyanin 색소(Park et al., 2015)는 유지의 자동산화 억제를 통한 지질과산화 반응 저해 효과(Bang et al., 1998), 혈중 콜레스테롤 저하를 통한 동맥경화 및 심혈관계 질환억제 효과(Lee, 2016), 혈중 지질을 낮추어주는 항 이상지질혈증 효과(Kim et al., 1999)가 보고되어 있다. 또한 생체내의 활성산소 제거 등과 같은 항산화 작용을 가져 뇌손상 뿐 아니라 각종 만성 퇴행성 질환 예방(Kim et al., 1999)과 염증반응에 중요하게 관여하는 매개인자인 nitric oxide, tumor necrosis factor α (TNF-α), interleukin1β (IL1β) 및 interleukin 6 (IL6)의 생성 억제효과를 보여 항염증(Han et al., 2005) 효과가 알려져 있어 인체에 무해한 천연색소 및 기능성 소재로 각광받고 있다. 이러한 뽕잎과 오디의 생리활성을 활용한 연구로는 뽕잎분말과 오디분말의 최적 혼합비율을 이용한 기능성 죽 제조(Kim et al., 2017), 뽕잎과 오디분말을 이용한 육류용 소스 제조 및 품질평가(Kim et al., 2016), 뽕잎두부 제조(Han et al., 2005), 뽕잎설기 제조(Lee, 2016), 뽕잎당면의 생리활성 평가(Kim et al., 2000), 뽕잎젤리 제조(Kim et al., 2006a), 뽕잎쿠키 제조(Lee, 2016), 뽕잎절편 제조(Kim et al., 2000), 오디젤리 제조(Kim et al., 2007, Moon et al., 2012), 오디요구르트 제조(Sung & Choi, 2014), 오디와인 제조(Son & Choi, 2013) 등이 있다. 이와 같이 뽕잎과 오디를 소재로 한 연구 대부분이 뽕잎과 오디 각각 단일물질로 이용한 연구들로 제한되어 있다. 묵은 우리나라 고유의 전통식품의 하나로 메밀, 도토리 및 녹두 등을 갈아서 물에 가라앉힌 앙금을 물과 함께 되직하게 쑤어 식혀서 굳힌 음식으로(Cho & Choi, 2007), 다른 재료를 첨가하기에 용이하고 열량이 낮은 장점이 있다(Choi, 2013). 따라서 본 연구에서는 뽕잎과 오디에 포함되어 있는 생리활성물질들 간의 시너지 효과를 이용하여 이너 뷰티 푸드를 제조하고자 뽕잎과 오디분말의 혼합비율을 달리하여 묵을 제조하고 그 품질특성을 평가하였다.

Methods

1. 실험 재료

본 실험에 사용한 동결건조 뽕잎과 오디분말은 (주)부안(Buan, Korea), 녹두 전분은 제일제당(CJ, Korea)에서 각각 구입하여 묵 제조 시 재료로 사용하였다.

2. 이너 뷰티 푸드(묵) 제조

뽕잎과 오디혼합분말을 녹두전분 대비 2%씩 첨가하여 제조한 묵의 recipe는 Table 1에 제시된 바와 같이 Figure 1의 제조방법에 의해 제조하였다. 묵을 제조하기 위해 시료 각각을 2 L용 내열성 파이렉스 용기(Pyrex, USA)에 혼합하여 3 h 동안 실온에서 침지한 후 induction cooker (Stovmon, Sparanise, Italy)를 사용하여 100℃에서 8 min, 80℃에서 20 min 동안 잘 저어가면서 가열 한 후 높이 4.5 cm×직경 13 cm의 원통용기에 부어 상온에서 1 h, 냉장고에서 2 h 굳힌 후 4 cm×4cm×3cm로 절단한 다음 색도, 기계 물성, 관능평가를 측정하였다. 이를 -70℃ 냉동고에서 동결 건조한 후 분말화하여 생리활성 분석용 시료로 사용하였다.

3. 색도 측정

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 색도측정(Yoo et al., 2002)은 색차계(Chroma Meter Cr-300; Minolta, Japan)를 사용하여 명도(lightness, L*), 적색도(redness, a*) 및 황색도(yellowness, b*)값으로 표시하였다. 각 시료 당 3회 반복 측정하여 그 평균값을 나타내었고, 표준 백색판(L*=96.69, a*=0.22, b*=1.89)을 사용하여 측정하였다.

4. 기계적 물성 측정

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 기계적 물성 측정을 위해 texture analyzer (TAXT Express V2.1, London, England)를 사용하여 texture profile analysis (TPA) test를 실시하였다. Probe는 직경이 36 mm, Trigger force는 5.0 g, pre-test speed, test speed는 1.0m, post-test speed 는 3.0 mm/s, distance는 8.0 mm으로 측정하여 경도(hardness), 탄력성(springiness), 검성(gumminess), 씹힙성(chewiness) 및 응집성(cohesiveness)을 측정하였다.

5. 일반성분 분석

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 일반성분은 AOAC 방법(AOAC, 2000)에 따라 수분은 105℃ 상압가열건조법, 조단백질은 semi-micro kjeldhal법, 조지방은 majonnier법, 조회분은 550℃ 직접회화법으로 측정하였다. 탄수화물 함량은 전체 성분 비율을 100%로 하여 수분, 조단백질, 조지방, 조회분 함량을 감한 것으로 산출하였다.

6. 무기질 함량 분석

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 무기질 함량은 AOAC 방법(AOAC, 2000)에 따라 습식 분해 후 Inductively Coupled Plasmaspectrometer (ICP, 7500a; Thermo Jarrell Ash, USA)를 사용하여 측정하였다. 즉, 조회분 함량은 시료 1 g을 0.5 N HNO3을 넣고 Whatman® glass microfiber filter (GF/C 90 mm; GE Healthcare, USA) 여과지로 여과 한 다음 0.5 N HNO3 50 mL로 정용하여 ICP로 분석하였다. 분석조건은 auxiliary gas flow 1.5 L/min, nebulizer pressure 30.1 psi, approximate RF power 950 W, frequency 27.12 MHz, type simultaneous, analysis pump rate 130 rpm, pump tibing type EP-19이었다.

7. 생리활성 물질 분석

1) 가바(γ-aminobutyric acid; GABA) 함량

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 GABA 함량은 Tomita et al. (2014)의 방법을 변형하여 실시하였다. 즉, 동결 건조한 시료분말 1 g에 증류수 40 mL을 가해 homogenizer (Polytron RT 2500E; Kinematica AG, Switzerland)로 2 min 균질화시켰다. 균질화한 시료는 4℃에서 10 min 원심분리(3,000 rpm) 한 후 상등액을 분리하고 침전물에 다시 증류수 40 mL를 가하여 2 min 추가 추출하였다. 추출물을 원심분리 하여 얻은 상등액을 모아 증류수를 이용하여 50 mL로 정용하였다. 추출물은 0.2 μm nylon syringe filter (Whatman, USA)로 여과하고 Nexera X2 system (Shimadzu, Japan)으로 분석하였다. 컬럼은 MightysilTM RP-18 GP column (4.6×250 mm, 5 μm; Kanto Chemical, Japan)을 사용하였고, 이동상 A는 HPLC water 2 L에 AccQ-Tag Eluent A concentrate 200 mL를 희석하였고 이동상 B는 AccQ-Tag Eluent B를 이용하였다. 용매 조성은 기울기 용리로 초기 A:B를 90:10으로 유지했으며, 15 min까지 80:20, 30 min까지 50:50, 50 min까지 0:100 및 60 min까지 90:10으로 분석하였다. 유속은 1.0 mL/min, 시료 주입량은 10 μL, 검출기는 fluorescence detector (Shimadzu)를 사용하여 Ex 250nm, Em 395 nm로 분석하였다.

2) DNJ 함량

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 DNJ 함량은 Hwang 등(Hwang et al., 2008)의 방법을 변형하여 측정하였다. 즉, 시료 1 g을 0.05 N HCl로 15 min 추출한 후 4℃에서 15 min 원심분리(3,000 rpm)하여 상등액을 추출용매 50 mL로 정용하여 0.2 μm nylon syringe filter (Whatman)로 여과시킨 후 FMOC-Cl로 DNJ 유도체화 시킨 후 측정하였다. HPLC [Nexera X2 system (Shimadzu)] 분석 조건은 컬럼은 Mightysil TM RP-18 GP column (4.6×250 mm, 5 μm, Kanto Chemical, Japan), 유속은 0.6 mL/min, 주입량은 10 μL, 검출기는 fluorescence detector를 사용하여 Ex. 254 nm, Em. 322 nm로 분석하였다.

3) Cyanidin-3-glucoside 함량

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 cyanidin-3-glucoside (C3G) 함량은 HPLC 방법으로 Ji 등(Ji et al., 2016)의 방법을 변형하여 실시하였다. 즉, 동결 건조한 시료분말 1 g에 추출용매(0.3% HCl을 함유한 80% 메탄올) 20 mL을 가하여 30 min 초음파 추출한 후 0℃에서 3,000 rpm의 속도로 15 min 원심분리하여 상등액을 취하고, 침전물에 추출용매 20 mL을 가해 추출하여 얻은 상등액을 추출용매 50 mL로 정용한 후 0.2 μm nylon syringe filter (Whatman, USA)로 여과하여 분석시료로 사용하였다. HPLC Nexera X2 system (Shimadzu) 분석조건은 컬럼은 Luna OmegaTM Polar C18 column (2.1×150 mm, 1.6 μm; Phenomenex, USA)을 사용하였고, 온도는 30℃, 시료 주입량은 2 μL, 유속은 0.3 mL/min, 검출기는 diode-array detector (Shimadzu)를 사용하였다.

8. 통계 처리

본 연구에서 얻은 모든 자료는 3회 반복 측정한 값을 이용하여 mean±standard deviation (M±SD)로 나타내었고, 유의성 검증은 Statistical Package for Social Sciences (SPSS, ver. 24.0; IBM, USA)를 이용하였으며, 시료간의 유의성 차이는 Duncan's multiple range test로 p<0.05 수준에서 검정을 실시하였다.

Results and Discussion

1. 색도

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 색도를 측정한 결과는 Table 2에 제시된 바와 같다. 명도(lightness)를 나타내는 L*값은 대조군 49.80에 비해 비교군인 ML0MF2 (뽕잎분말 0 g, 오디분말 2 g) 34.71, ML0.5MF1.5 (뽕잎분말 0.5 g, 오디분말 1.5 g) 33.43, ML1MF1 (뽕잎분말 1 g, 오디분말 1 g) 32.54, ML1.5MF0.5 (뽕잎분말 1.5 g, 오디분말 0.5 g) 35.85, ML2MF0 (뽕잎분말 2 g, 오디분말 0 g) 33.83으로 모두 유의하게 낮게 나타났다. 특히 뽕잎과 오디분말 1:1 비율로 2% 첨가된 ML1MF1의 색도가 대조군에 비해 가장 낮게 나타났다. 이는 서로 보색관계에 있는 오디의 붉은색과 뽕잎의 초록색이 혼합된 결과로 사료된다. 적색도(redness)를 나타내는 a*값은 대조군 0.55, 비교군 ML0MF2 3.94, ML0.5MF1.5 2.63, ML1MF1 -0.76, ML1.5MF0.5 -4.82, ML2MF0 -5.38로 오디분말만 첨가된 ML0MF2가 3.94로 가장 높게 나타났다. 이는 오디의 안토시안 색소의 영향으로 사료된다. 황색도(yellowness)를 나타내는 b*값은 대조군 -4.20에 비해 비교군 ML0MF2-5.25, ML0.5MF1.5-0.77, ML1MF1 2.03, ML1.5MF0.5 5.17, ML2MF0 8.10으로 뽕잎분말만 첨가되었을 때 가장 높았다. 이는 뽕잎의 녹색색소의 영향으로 사료된다. 본 실험의 이러한 결과는 뽕잎분말의 첨가량이 증가할수록 L*, a*값은 낮아지고, b*값은 높아진다고 보고한 뽕잎젤리(Kim et al., 2006a) 연구와 유사한 경향이었다. 또한 오디분말의 첨가량이 증가할수록 L*, b*값은 낮아지고, a*값은 높아졌다고 보고한 뽕잎과 오디분말을 이용한 육류용 소스(Kim et al., 2016)의 연구와도 비슷한 경향을 나타내었다.

2. 기계적 물성

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 기계적 물성으로 경도, 탄력성, 씹힘성, 검성 및 응집성을 측정하여 Table 3에 제시하였다. 경도는 대조군 127.6에 비해 비교군 ML0MF2 126.07, ML0.5MF1.5 117.60, ML1MF1 109.77, ML1.5MF0.5 97.8, ML2MF0 87.27로 모두 낮았으며, 특히 뽕잎분말만 첨가되었을 때 가장 낮았고 오디분말만 첨가되었을 때 가장 높았다. 탄력성은 대조군 0.91, 비교군 ML0MF2 0.91, ML0.5MF1.5 1.01, ML1MF1 0.94, ML1.5MF0.5 0.87, ML2MF0 0.90으로 대조군과 비슷하게 나타났다. 검성은 대조군 56.85, 비교군 ML0MF2 60.58, ML0.5MF1.5 51.29, ML1MF1 48.47, ML1.5MF0.5 47.17, ML2MF0 50.54로 ML0MF2가 가장 높았으며 ML1.5MF0.5에서 가장 낮았다. 씹힘성은 대조군 51.56, 비교군 ML0MF2 55.11, ML0.5MF1.5 52.24, ML1MF1 45.31, ML1.5MF0.5 40.82, ML2MF0 45.47로 오디분말만 첨가되었을 때 가장 높았다. 이는 오디 젤리의 이화학적 특성연구에서 오디 분말 함량이 많은 젤리에서 검성이 높았다는 연구결과와 비슷한 경향을 나타내었다(Kim et al., 2016).

3. 일반성분

뽕잎과 오디 혼합분말을 녹두전분 대비 2%씩 첨가하여 제조한 묵의 일반성분 분석 결과는 Table 4에 제시된 바와 같다. 수분함량은 대조군은 1.30%, ML0MF2 1.02%, ML0.5MF1.5 0.63%, ML1MF1 1.30%, ML1.5MF0.5 1.26%, ML2MF0 1.00%로 비교군이 대조군보다 낮은 함량이었으나 ML1MF1군은 1.30%로 대조군과 같게 나타났다. 조지방은 대조군 0.30%에 비해 ML0MF2 1.19%, ML0.5MF1.5 1.51, ML1MF1 1.12%, ML1.5MF0.5 1.41%, ML2MF0 1.07%로 비교군이 유의하게 높게 나타났다. 조단백질은 대조군 1.92%에 비해 비교군 ML0MF2 4.62%, ML0.5MF1.5 2.52%, ML1MF1 4.44%, ML1.5MF0.5 2.64%, ML2MF0 4.32%로 ML0MF2이 가장 높고 ML1MF1이 2번째 ML0.5MF1.5가 가장 낮게 나타났다. 조회분 함량의 경우 대조군 2.11%에 비해 ML0MF2 2.w94%, ML0.5MF1.5 3.21%, ML1MF1 3.06%, ML1.5MF05 2.87%, ML2MF0 2.83%으로 나타났다. 탄수화물 함량은 대조군 94.36%, ML0MF2 90.23%, ML0.5MF1.5 92.12%, ML1MF1 90.08%, ML1.5MF05 91.83%, ML2MF0 90.78%로 비교군이 낮게 나타났다. 이는 오디 착즙액 첨가비율에 따른 품질 특성에서 오디 착즙액의 첨가량이 증가할수록 조단백질의 함량이 증가하였다고 한 결과(Kim et al., 2007)와 비슷한 경향이었다. 또한 Kim et al. (2016)의 오디 분말을 이용한 젤리제조 및 이화학적 특성에 관한 연구와 누에 분말을 이용한 젤리(Kim et al., 2006b)와 뽕잎을 이용한 젤리(Kim et al., 2006a)에서 오디, 누에 및 뽕잎분말 첨가량이 증가할수록 조단백질의 함량이 증가되었다는 보고와도 같은 양상이었다.

4. 무기질 함량

뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 무기질 함량분석 결과는 Table 5에 제시된 바와 같다. 칼슘함량은 대조군 35.11 mg/100 g에 비해 ML0MF2 42.50 mg/100 g, ML0.5MF1.5 107.68 mg/100 g, ML1MF1 93.20 mg/100 g, ML1.5MF0.5 72.77 mg/100g, ML2MF0 71.27 mg/100 g으로 비교군이 모두 높았다. 칼륨 함량도 대조군 12.65 mg/100 g에 비해 ML0MF2 36.59 mg/100 g, ML0.5MF1.5 58.92 mg/100 g, ML1MF1 50.53 mg/100 g, ML1.5MF0.5 42.79 mg/100g, ML2MF0 43.34 mg/100 g으로 비교군이 높게 나타났다. 마그네슘 함량도 대조군 12.43 mg/100 g에 비해 ML0MF2 14.15 mg/100 g, ML0.5MF1.5 33.29 mg/100 g, ML1MF1 27.95 mg/100 g, ML1.5MF0.5 22.60 mg/100g, ML2MF0 21.69 mg/100 g으로 비교군이 높게 나타났다. 즉, 칼슘, 칼륨, 마그네슘 함량은 ML0.5MF1.5가 가장 높았고 ML1MF1이 2번째로 높았고 ML0MF2가 가장 낮았다. 뽕잎분말의 칼슘 함량은 2517.02 mg/100 g, 칼륨은 2107.04 mg/100 g, 마그네슘은 467.39 mg/100 g으로 오디분말의 칼슘 216.55 mg/100 g, 칼륨 1176.05 mg/100 g, 마그네슘 91.65 mg/100 g에 비해 각각 약 10배, 2배, 5배 가량 높았다. 이 같은 결과로 볼 때 혈압조절 기능(Shin et al., 2014)이 있는 칼슘, 칼륨, 마그네슘은 뽕잎분말이 오디분말에 비해 더 많이 함유되어 있었다. 그러나 이를 혼합하여 묵을 제조하였을 때는 꼭 같은 경향을 나타내지는 않았다. Kim et al. (2006a)의 뽕잎 젤리에서 칼슘, 칼륨, 마그네슘 함량이 뽕잎과 오디분말 첨가수준이 높아질수록 대조군에 비해 증가하였다는 결과와는 유사한 경향이었다.

5. 생리활성 물질

1) 가바 함량

가바(γ-aminobutyric acid; GABA)는 뇌 조직 혈류의 흐름 촉진으로 뇌세포의 대사기능을 촉진시켜 기억력을 증진시키는 생리활성 물질이다(Kim et al., 2007). 또한 가바는 여러 인체조직에서 glutamate decarboxylase에 의해 glutamic acid로부터 생성되어 혈압이 올라가는 것을 억제하는 항고혈압성 생리활성 물질로도 알려져 있다(Kim et al., 1999). 이에 본 연구에서는 뽕잎에 포함된 가바의 효능을 활용하여 이너 뷰티 푸드를 개발하고자 뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 가바 함량을 측정하였으며 그 결과는 Table 6에 제시하였다. 대조군은 0.03 mg/100 g, 비교군인 ML0MF2 1.34 mg/100 g, ML0.5MF1.5 2.73 mg/100 g, ML1MF1 2.84 mg/100 g, ML1.5MF0.5 2.95 mg/100g, ML2MF0 3.49 mg/100 g으로 뽕잎분말만 2% 첨가하였을 때 가장 높았고 ML1MF1가 3번째로 높았으며 오디분말만 첨가하였을 때 가장 낮았다. 뽕잎분말의 가바 함량은 122.23 mg/100 g으로 오디분말의 7.67 mg/100 g에 비해 약 16배 많은 양이 함유되어 있어 뽕잎분말만 2% 첨가한 묵에서 가장 높게 나타난 것으로 판단된다. 즉, 가바가 주로 뽕잎에 포함되어 있기 때문이며 이러한 가바는 비단백질 구성 아미노산으로 글루탐산 탈탄산효소(glutamate decarboxylase, GAD)의 작용에 의해 글루탐산이 탈탄산 되어 생합성 되는 물질로(Yoo et al., 2002), 혈압 상승 억제제이며 인체 내의 신경계나 혈액에 함유되어 생리기능에 중요한 물질이다. 그러므로 가바가 함유된 뽕잎을 오디와 혼합하면 고혈압을 예방하는 기능성식품 소재로 활용가능성이 높을 것이다.

2) DNJ 함량

1-deoxynojirimycin (DNJ)는 α-glucosidase에 대해 저해활성을 갖는 물질로 혈당강하 효과가 있는 것으로 알려져 있다(Shin et al., 2014). 뽕잎에 포함된 DNJ의 효과를 활용하고자 뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 DNJ 함량을 측정하였으며 그 결과는 Table 6에 제시된 바와 같다. 대조군에서는 DNJ 함량이 측정되지 않았으나 ML0MF2 1.30 mg/100 g, ML0.5MF1.5 3.49 mg/100 g, ML1MF1 3.75 mg/100 g, ML1.5MF0.5 4.13 mg/100 g, ML2MF0 5.28 mg/100 g으로 뽕잎분말만 첨가하였을 때 가장 높고 ML1MF1가 3번째로 높았으며 오디분말만 첨가하였을 때 가장 낮았다. 이는 DNJ가 뽕잎분말 268.89 mg/100 g으로 오디분말 42.74 mg/100 g에 비해 6.3배 높게 함유되어 있기 때문에 뽕잎분말만 2% 첨가한 묵에서 높게 나타난 것으로 판단된다. 뽕잎에 함유된 DNJ와 같은 알칼로이드 성분들은 소장내 α-glucosidase를 저해하여 당흡수를 억제하는 것으로 알려져 있으므로(Son & Choi, 2013) 이러한 성분이 함유된 뽕잎과 오디분말을 함께 섞어 제품을 제조하면 혈당 및 지질대사 등의 당뇨증세의 개선에 효과적일 것으로 생각된다.

3) Anthocyanin 과 Cyanidin-3-glucoside (C3G) 함량

안토시아닌은 플라보노이드에 속하는 대표적인 천연색소로 포도, 오디, 블루베리, 검정콩 및 흑미에 다량 존재하며, 항산화, 항염, 신경보호 등의 생리활성 작용을 한다(Yoo et al., 2002). 특히 C3G는 오디의 천연색소 중 가장 안정한 형태로 분리, 정제가 용이하며 항산화 작용이 높은 기능성 소재로 각광받고 있다(Kim et al., 2017). 이에 본 연구에서는 오디에 포함된 C3G를 활용하고자 뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 제조한 묵의 안토시아닌과 C3G 함량을 측정하여 Table 6에 제시하였다. 대조군에서는 안토시아닌이 측정되지 않았으며 ML0MF2 75.41 mg/100 g, ML0.5MF1.5 29.01 mg/100 g, ML1MF1 19.88 mg/100 g, ML1.5MF0.5 18.51 mg/100 g, ML2MF0 1.97 mg/100 g, 뽕잎분말 은 7.21 g/100 g, 오디분말은 5216.69 mg/100 g으로 오디 분말이 뽕잎분말 에 비해 약 723배 가량 많이 함유되어 있었으며, 이로 인해 오디분말만 2% 함유된 ML0MF2에서 가장 높게 나타났으며 뽕잎과 오디분말을 동량으로 혼합하여 첨가해 제조한 ML1MF1가 3번째로 높게 나타났다. C3G는 대조군에서는 측정되지 않았으며 ML0MF2 95.48 mg/100 g, ML0.5MF1.5 40.48 mg/100 g, ML1MF1 21.08 mg/100 g, ML1.5MF0.5 11.63 mg/100 g, ML2MF0 0.10 mg/100 g으로, 뽕잎분말은 3.15 g/100 g, 오디 분말은 3584.16 mg/100 g으로 오디 분말의 C3G 함량이 뽕잎분말에 비해 약 1138배 가량 많이 함유되어 있어 오디 분말만 2% 첨가된 ML0MF2에서 그 함량이 가장 많았다. 즉, C3G가 오디에 많이 함유되어 있으며(Son & Choi, 2013) 이러한 C3G는 강력한 항산화력을 갖고 있어 활성산소 생성 억제 등을 통해 각종 질환예방 기능성 식품과 이너 뷰티 푸드 개발에 활용될 수 있는 소재로 기대된다.

Conclusion

이상의 결과, 뽕잎과 오디 혼합분말을 녹두 전분 대비 2% (w/w) 첨가하여 묵을 제조하였을 때 조단백질은 오디 분말만 첨가된 ML0MF2가, 칼슘과 칼륨, 마그네슘 함량은 ML0.5MF1.5이 가장 높았다. GABA와 DNJ는 뽕잎분말만 2% 첨가하였을 때 가장 높았으며 안토시아닌과 C3G는 오디 분말만 2% 첨가하였을 때 가장 높았다. 이 같은 결과로 볼 때 생리활성물질인 GABA와 DNJ 함량을 높게 한 제품을 제조하기 위해서는 뽕잎분말을 많이 첨가하는 것이 긍정적이다. 항산화 기능을 강화시키는 이너 뷰티푸드나 제품을 제조하기 위해서는 안토시아닌과 C3G 함량이 많이 함유되어 있는 오디 분말 함량을 높여 제조하는 것이 바람직하다. 그러므로 단백질 함량을 증가시키고 뽕잎에 많은 GABA, DNJ와 오디에 많은 안토시아닌과 C3G 두 가지 물질을 혼합하여 시너지 효과를 내기 위해서는 뽕잎분말과 오디 분말을 1:1로 하는 것이 바람직하다고 판단된다. 본 연구 결과는 향후 뽕잎과 오디를 활용한 이너 뷰티 푸드와 건강식으로의 활용 가능성이 높을 것으로 생각된다.
