Comparative study about immunity modulation on IgE biosynthesis of Phellinus linteus (PL), Germinated Brown Rice (BR) and Phellinus linteus Grown on Germinated Brown Rice (PB)

ABSTRACT
Background and objectives: To compare the immunomodulatory effects on the amount of IgE synthesis, the distribution of T-cells and the amount of cytokine secretion in the MLN (Mesenteric Lymph Node) between Phellinus linteus (PL), Germinated Brown Rice (BR) and Phellinus linteus Grown on Germinated Brown Rice (PB) and thereby to examine their relationships with atopic dermatitis. Materials and methods: All the extracts of PL, BR and PB were dissolved in the propylene glycol. Then, male mice aged 4 weeks were divided into four experimental groups, each of which comprised ten mice. These mice were provided with experimental foods in a certain environment and then orally given each extract. Through a retro-orbital bleeding, the peripheral blood was sampled. This blood sample was used for an analysis of IgE.
Results:
The concentration of serum IgE was significantly decreased in the experimental groups where PL, BR or PB were administered as compared with the control group (Fig, 1-1). Besides, the concentration of serum IgE was also significantly decreased in the experimental groups where PL, BR or PB were administered in the MLN as compared with the control group (Fig 1-1). The number of CD4+ T-cells was significantly greater in the experimental groups where PL and PB were administered as compared with the control group. But the number of CD8+ T-cells was the smallest in the experimental group where PL was administered(Table 1). Besides, of all the extracts, BR had an effectiveness in enhancing the concentration of IFN-γ (Interferon-γ) to the greatest extent in the MLN (Fig, 1-2).
Conclusions:
It has been reported that Th2 cytokines and serum IgE are relatively higher in more than 80% of patients with atopic dermatitis. Our results showed that PB reduced the amount of the secretion of Th2 cytokine. Thus, by modulating the amount of IgE synthesis, it had an immunosuppressive effect on various types of inflammatory markers. It is also presumed that PB might reduce the amount of the secretion of pro-inflammatory cytokines in MLN. It can therefore be inferred that PB might also have a treatment effect on atopic dermatitis. Further studies are warranted to examine the relationship between PB and atopic dermatitis.
