Anti-inflammatory Effect of Cornus officinalis Fruit Extract for Potential of Cosmetic Ingredient

ABSTRACT
Cornus officianalis has been used in oriental medicine and has been reported to have many functions such as reduction of melanin production, anti-cancer, antibacterial function, and etc.. In this study, we investigated the reduction of pro-inflammatory factor as a functional ingredient for cosmetic products from the 80% ethanol extract of Cornus officianalis fruit (CEE). We performed MTT assay for cell viability. Major inflammatory factors was measured by nitric oxide (NO) inhibition assay, immunoassay, and reverse transcription factor (RT-PCR). The cytotoxicity of CEE did not appear in the range of 125 μg/mL, 250 μg/mL, 500 μg/mL on RAW 264.7 cells. Inflammatory response on RAW 264.7 cells were induced by lipopolysaccharide (LPS), followed by CEE treatment at indicated concentrations (0 μg/mL, 125 μg/mL, 250 μg/mL, and 500 μg/mL). The CEE showed inhibitory effect on production of NO by 5.9%, 21.3%, and 34% at 125 μg/mL, 250 μg/mL, and 500 μg/mL concentrations. In addition, production of prostaglandin E2 (PGE2) was decreased by approximately 3%, 15%, and 39%, upon the CEE treatment at concentrations of 125 μg/mL, 250 μg/mL, and 500 μg/mL respectively. The production of interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α were decreased by approximately 57%, 52% and 34%, respectively upon CEE treatment at 500 μg/mL. Also, CEE showed inhibitory effects on the expression of IL-1β, IL-6, and TNF-α genes on LPS-induced RAW 264.7 cells. From the results above, we conclude that CEE indicated significant anti-inflammatory effects. And these results will be a basis for the development of cosmetology in skin care and improvement of skin problem.
